RESUMO

The immunosuppressant effects of the antineoplastic piperazine drugs spirobramine and prospidin in respective doses of 500 and 600 mg/kg (1/2 of the LD50) were studied in experiments on cats. Spirobramine was shown to exert a more pronounced immunoinhibitory action on the primary and secondary immune response induced by sheep red blood cells.

The morpho-functional state of the muscle lymphoid tissue of mice treated with karmionmycin (LD50, 1.1 mg/kg) was studied experimentally. Development of a number of changes evident of the cell disorganization in the lymphoid tissue was shown. The cytological shifts in the thymus and spleen were reversible, while the destructive period in the strumous gland was more prolonged. The morpho-structural normalization of the spleen did not coincide with reduction of its immunological function. The studies on the karminomycin effect on the kinetics of the primary immune response showed dependence of the antibiotic effect on the temporal ratio between the antigenic stimulus and the drug administration. The antibiotic had no effect on the inductive phase of the primary immune response but significantly suppressed its productive phase. On reproduction of the secondary immune response, a decrease in the content of the antibody-forming cells in the spleen was observed, production of the circulating antibodies remaining unchanged.

Methotrexate administered to albino mice in a dose of 0.5 mg/kg for a period of 10 days caused significant (two period) changes in the central and peripheral immunocompetent tissue. During the first--destructive--period there occurred a marked reduction in the weight of the spleen and the thymus, a cut in the count of the nuclear cells in them, and also a significant depletion of the cellular composition of the lymphoid organs. Suspension of the cytostatic was followed by a gradual restoration of all the indices under study, slower in the thymus than in the spleen. In the animals immunized with the tetanus toxoid against the background of the methotrexate administration all the changes in the lymphoid organs developed under a preponderant influence of the cytostatic agent; the action of the antigen was depressed during the whole experiment. Specific antibodies appeared in the blood of these animals later and their titres were lower than in control; experimental mice were also less resistant to tetanus toxin.

The leucocyte counts in the peripheral blood and titers of the serum lysozyme under conditions of various experimental models were compared. Albino rats, mice, guinea pigs and rabbits exposed to the effect of various factors of a biological and chemical nature were used in the experiments. In addition, observations on humans immunized with bacterial and viral vaccines were analyzed. Development of 3 reaction types was shown to be possible. In the 1st type the dynamics of the lysosome activity and blood leucocyte counts changed in the same way (immunization with smallpox vaccine, listeria infection). Decreased counts of the leucocytes due to the use of cytostatic drugs were accompanied by a parallel decrease in the lysozyme activity. Such variant of the reaction was predominating. The 2nd type of the shifts characterized by an impaired synchronous pattern of the dynamics of the indices was not so frequent (the use of high doses of cytostatics). The 3rd type of the reactions with an exactly opposite character of the shifts in the leucocyte counts and lysozyme activity was observed only as an exclusion. It is concluded that the quantitative dynamics of the leucocytes is not enough by itself for creating an opinion on the nature of the changes in the serum lysozyme. The possible role of macrophages in the above processes is discussed.
